Simply, specific structure of proteins determines specific interaction with substrates, thus role of protein conformation very important to be able to get correct three dimensional structure and to form active domain of enzyme.
For every singly, unique reaction, there might be only few possible interaction, structure to be able to interact and react with desired, specific molecule or substrate, thus wrong conformation, changing conformation changes enzyme specificity for specific molecule, it makes less specific if it differs from very specific structure. And some enzymes might have group specificity so that they can react with a group of similar molecules.
